ABSTRACT

@#[Abstract] Objective: To detect the expression of CD39 in head and neck squamous cell carcinoma (HNSCC) tisseus, and to analyze its correlation with patients’clinicopathological features and its prognostic significance. Methods: Tissue specimens and case data of 85 patients with HNSCC underwent surgery at Cancer Hospital of Tianjin from May 2012 to December 2013 were collected for this study. Gene chips were obtained from Oncomine database, and HNSCC cell lines SCC15, UM1, and Cal25 were selected for this study. Online analysis was performed to compare the differential expression of CD39 in buccal mucosa (BM) tissues and HNSCC tissues, Western blotting and Immunohistochemistry (IHC) were used to detect the protein expression of CD39 in HNSCC tissues. Spearman’ s correlation analysis was used to study the correlation between the expressions of CD39 and clinicopathological features of HNSCC patients. Both Kaplan-Meier curve analysis and Log rank test were used to analyze the association between the expression of CD39 in HNSCC tissues and the survival of patients, and Cox risk proportional regression model was used to evaluate the relationship between CD39 expression and the risk of relapse. Results: The transcription level of CD39 was obviously up-regulated in HNSCC tissues than in BM tissues (P<0.01), and CD39 expression was detected in HNSCC cell lines SCC15, UM1 and Cal25. Dexamethasone (DXM) could enhance the expression of CD39 in UM1 cells in dose-dependent manner. CD39 was highly expressed in 53 (62.4%) HNSCC patients, which was positively correlated with preoperative chemotherapy (r=0.234, P<0.05). The recurrence-free survival (RFS) of patients with high CD39 expression was significantly shortened (P<0.05), and high CD39 expression was an independent relapse risk factor (HR=2.328, 95%CI=1.091-4.967; P<0.05) for patients with HNSCC. Conclusion: CD39 is DXM-inducively and constitutively expressed in HNSCC. And over-expression of CD39 is an independent predictor of poor prognosis in HNSCC patients, indicating its important role in the progression of HNSCC.

@#肿瘤浸润性免疫细胞(TIC)参与构成肿瘤免疫微环境,调控肿瘤的生长,并影响患者的生存及抗肿瘤治疗的疗效。 TIC分布模式的类型主要有冷肿瘤型、混合型和间质型。这3种分布类型在免疫细胞的种类、数量与比例和分布位置上呈现明显 的差别。在不同肿瘤、不同个体,甚至同一肿瘤的不同区域之间,TIC分布模式既有差异性也有规律性,既有模式相似但功能相异 的差异性,也有模式不同但功能相近的规律性。TIC分布模式的分型体现的就是其差异性。近来研究发现,利用TIC分布模式可 以预测患者的预后及抗肿瘤治疗的疗效,这体现的就是TIC分布模式的规律性。本文就该领域的研究进展进行综述。

ABSTRACT

Objective:To investigate the function and mechanism of cyclin-dependent kinase 4/6 (CDK4/6) inhibitor PD0332991 on clo-nogenicity of mantle cell lymphoma (MCL) cells. Methods:The effect of PD0332991 on MCL cell cycle distribution was assessed by flow cytometry;Western blot was used to test expression level of Rb protein and phosphorylated Rb protein in MCL after treatment with PD0332991;colony forming assay was performed to test the role of PD0332991 and mitoxantrone and their combination on colo-ny forming activity in MCL. Results:Flow cytometry revealed that PD0332991 can increase G0/G1 phase MCL cells and significantly de-crease S phase cells, leading to G0/G1 cell arrest. Western blot confirmed that PD0332991 exerted no effect on Rb protein expression but suppressed levels of phosphorylated Rb protein. Colony forming assay showed that PD0332991 significantly suppressed colony for-mation and enhanced the effect of mitoxantrone on colony forming activity in MCL. Conclusion:This study revealed that CDK4/6 inhib-itor PD0332991 induced G0/G1 cell arrest and increased the effect of mitoxantrone on MCL clonogenicity by suppressing levels of phosphorylated Rb protein.

ABSTRACT

The aims of the present study were to describe an impression method of "inner circular sealing area" and to evaluate the effect of the method on retention, aesthetics and comfort of complete dentures, which lack labial base for patients with maxillary protrusions. Three patients were subjected to the experiment, and two sets of complete maxillary dentures were made for each patient; the first set was made without labial base via an inner circular sealing area method (experimental group) and the second had an intact base that was made with conventional methods (control group). Retention force tests were implemented with a tensile strength assessment device to assess the retention and a visual analogue scale (VAS) was used to evaluate the comfort between the two groups. Results showed larger retention force, better aesthetics and more comfort in the experimental group. The improved two-step impression method formed an inner circular sealing area that prevented damage to the peripheral border seal effect of the denture caused by incomplete bases and obtained better denture retention.


O objetivo deste caso foi descrever um método de impressão por "área de selamento circular interno" e avaliar o efeito deste método na retenção, estética e conforto de próteses totais sem base labial para pacientes com protrusão maxilar. Três pacientes foram objeto desta experiência e foram feitas duas próteses maxilares completas para cada um deles; a primeira foi elaborada sem base labial pelo método de área de selamento circular interno (grupo experimental) e a outra teve uma base feita pelo método convencional (grupo controle). Foram realizados testes de retenção com estudo de tensão para avaliar a retenção e para avaliação do conforto dos dois grupos, foi empregada a escala analógica visual (EAV). Os resultados demonstraram que o grupo experimental apresentou força de retenção maior, estética melhor e mais conforto. O método modificado de impressão em duas etapas formou uma área de selamento circular interno que evitou danos ao selamento periférico causados por bases incompletas e obteve melhor retenção da prótese.
